I was told by one of my friends that I should run a little pre-mix in my tank for the first 1/4 tank or so to make sure my oil pump is working. Is this true or is it something he made up?

Oil pumps typically over deliver when they go bad. If you've recently messed with the lines and feel you may have air in them, you could run premix but if everything was working properly last year I see no reason to do it.

There is a older gentleman in town here that has a 95 or 96 vmax 600 twin with over 10,000 miles on it never been apart and he told me that he always premixes the first tank of fuel in it every year. MUST BE DOIN SOMETHING RIGHT!!!

No need to pre-mix unless you took the lines apart and didn't get them completely bled of all air. On some of the older sleds the lines would let air in over the summer and pre-mixing was a way to offset this. If you are worried just make sure your lines and pump do not have any air in them.
